Úspěch však představuje výzvu díky fyziologickým bariérám trávicího traktu. Pokud zvýšení rozpustnosti léčiva předchází cílení, lze rozpouštění, jako krok limitující rychlost, obejít, urychlit vstřebání léčiva a zvýšit jeho perorální biodostupnost. Pro optimalizaci dávky a terapeutického efektu je kromě toho potřebné zadržet přípravek v místě cílení. Cílem této práce je proto vývoj systému pro doručení a cílení omezeně rozpustného léčiva cyklosporinu A (CyA) do kolonu. V první části experimentu byly využity preformulační studie k dosažení zvýšené rozpustnosti a zrychleného uvolnění CyA pomocí tzv. liquisolid sytému (LSS) nebo přípravou interaktivních směsí společným mletím (komletím). Probiotics and their importance in human nutrition

A great deal of immune system is located in the colon thanks to a large number of microbs that influence not only digestive tract of a human. The contact of these microbs with the digestive tract of a human is very important for other development of immune defensive mechanisms and the natural immune response, which supports health of a host. That is why in recent years a great deal of attention and efforts have been paid to possibility of human health status improving by a modulation of the intestinal microflora. As probiotics are indicated living microorganisms, that can have positive effects on human health and some species, for example Saccharomyces boulardii, have positive effects in prevention and treatment of many diseases associated with diarrhea. The company Valosun a.s. has provided for testing the probiotic food supplement which is suitable in acute diarrheal disorders. This product has been tried by patients of gastroenterology ambulance. The aim of sociological survey was to map knowledge of respondents in the field of probiotics and probiotic food supplements.

This Bachelor thesis explains the basics of biochip or microarray data interpretation, starting with short introduction to genetics, especially genetic information significance evaluating. The focus was set mainly on the set of scripts transforming and analyzing the sample data. The data used in this thesis are a result of biochip analysis of the Colon Tumor tissues. The secondary result represents disclosing the main marker for this particular type of cancer, the primary result is evaluation of marker significance in the context of signaling pathways. The resulting pathways are sorted by relevance.

v Abstract Author: Jan Pavlíček Title: A case report of physiotherapeutic treatment provided to a patient following liver transplantation and colon resection Objectives: This bachelor thesis aimed to introduce theoretical information concerning issues with a liver transplant, and large intestine resection and elaborate on a case report of a patient that underwent the above-mentioned procedures. Methods: This bachelor's thesis is divided into two parts. The liver and large intestine's anatomy, physiology, and pathophysiology are discussed in the first general section. Following chapters are devoted to the main treatment options for advanced liver diseases, i.e. liver transplantation, and indication and possibilities of colon resection. Physiotherapeutic approaches used in the mentioned procedures and further options of treatment for these patients are mentioned in the later chapters. The second specialized part describes a case report of a patient post-liver transplantation and large intestine resection. Included is a report of therapies that the patient underwent as part of ongoing professional practice in IKEM in January and the beginning of February 2023. Histological examination of the colon

This bachelor´s thesis deals with histological examination of the colon, namely the laboratory diagnosis of colorectal cancer. Colorectal cancer is a highly relevant issue today not only in the Czech Republic, not least because of its continually growing incidence worldwide. Laboratory analysis of this disease is not only important from the aspect of diagnosis per se. Determining the type and invasive stage of the disease is also crucial for the therapeutic strategy in a given patient. Immunohistochemistry testing of the colon also aids in the diagnosis of Lynch syndrome, a hereditary disease with a high risk of colorectal cancer. The first part of the thesis is theoretical and aims to introduce not only the subject of colorectal cancer but also the anatomical, histological and pathological features of the colon. This section also describes the methods used to obtain tissue samples from the colon and to process these in a histopathology laboratory. This information regarding the histopathological processing of colon tissue is then applied and described in detail in the methodology section of the thesis. This describes the process of routine staining methods such as haematoxylin-eosin and Alcian Blue. It also details the manually performed immunohistochemical methods and related reagents, including the use of antibodies. The next section presents a statistical analysis of a sample of patients with colorectal cancer treated at the Jihlava Hospital between 2018 and 2019. These data confirm the generally accepted fact that there is a higher incidence of colorectal cancer in men and that this cancer predominantly affects patients aged between 60 to 70 years. Furthermore, this work shows that defects in the MMR proteins associated with Lynch syndrome are not detected in even one half of the patients tested.

Objectives: The aim of this bachelor thesis is a detailed elaboration of a case study of physiotherapeutic treatment of a patient after proctocolectomy with temporary ileostomy and summary of theoretical knowledge related to this diagnosis, its causes, consequences and rehabilitation treatment options. This thesis was created within a continuous professional practice at the Institute of Clinical and Experimental of Medicine in Prague at the KARIP Resuscitation Department in the period 14/01/2022 - 04/02/2022. Methods: The main division of this work is a theoretical part and a special part. The theoretical part will contain brief anatomical and physiological information on the small and large intestine. Next, I will briefly mention the types of intestinal polyposes and focus in detail on familial adenomatous polyposis. In the special part, there will be processed a case study of a physiotherapeutic treatment of the patient after proctocolectomy with temporary ileostomy together with records of individual therapies. Role of intestinal circadian clock in epithelial transport, proliferation, and tumourigenesis

AABBSSTTRRAACCTT The molecular circadian clock enables anticipation of environmental changes. In mammals, clocks are ubiquitously present in almost all tissues and they are comprised of transcriptional-translational feedback loops of the so-called clock genes. The central clock represents the intrinsic pacemaker which is located in suprachiasmatic nuclei (SCN) of hypothalamus and synchronizes peripheral clocks. Clockwork system in alimentary tract and its regulatory link to intestinal functions are poorly understood. Therefore the objective of the thesis was to characterize molecular clock in particular parts of the rat intestine and to elucidate its link to the intestinal transport, regulation of cell cycle and neoplastic transformation in colonic tissue. We used quantitative RT-PCR (qPCR) to determine circadian profiles of mRNA expression of clock genes in the epithelium of duodenum, jejunum, ileum, and colon of rat. Furthermore, we analysed the expression of genes coding sodium chloride transporters and channels as well as cell cycle regulators in colon. To focus more precisely on different structures of intestinal epithelia we used laser capture microdissection. Study of genetic factors modifying the risk of onset and progression of colorectal and pancreatic cancer

Introduction: The aim of this study was to evaluate the role of genetic and lifestyle factors in the risk of onset and progression of colorectal and pancreatic cancer. The first part deals with the etiological factors and the importance of polymorphisms in biotransformation enzymes and genetic alterations in the gene CHEK2 in the origin of these malignancies. In the second part, the ABC transporter genes were analyzed as potential prognostic and predictive markers of a treatment's outcome. Materials and methods: The polymorphisms and other genetic alterations were detected using real-time PCR, allelespecific PCR and PCR-RFLP methods in DNA which was extracted from the blood of patients. The frequency of polymorphisms was evaluated and their importance was assessed with regard to the available epidemiological data. Gene expressions were determined by qPCR in paired samples of tumor tissue and adjacent non-tumorous parenchyma. Results: A majority of the observed polymorphisms failed to show a relationship between their presence and the risk of any of these malignancies. CYP2A13 variant allele*7 coding inactive enzyme was found in 7 of 265 controls and in none of 235 pancreatic carcinoma patients.
